@EN A new bioluminescent assay method for the activity of phospholipase A2(PLA2; phosphatitylcholine 2-acylhydrolase: EC 3.1.1.4) was developed using bioluminescent marine bacteria. A dark mutant of Vibrio harueyi (designated M-17), the in vivo
light
intensity of which was stimulated up to one thousand fold in the presence of myristic acid, was used for this assay. Sn-1-Stearoyl-sn-2myristoryl choline and bovine pancreas phospholipase A2 were used in this assay system, and produced myristic
acid was
quantified using V. harveyi mutant M-17. Activity was detected in less than one min with as little as 5 mU of phospholipase A2 and 5 nM of myristic acid production.
